DETERMINATION OF QUERCETIN LEVEL IN BAWANG DAYAK (Eleutherine americana Merr) WITH HIGH PERFORMANCE LIQUID CHROMATOGRAPHY (HPLC) METHOD Ester Melurin Filadelfia NIM: 1843002 ABSTRACT Bawang dayak are one of the plants that are widely used as herbal medicine ingredients that treat many diseases. Quercetin is a flavonoid group that has antioxidant activity and is found in this plant. This study aims to determine the content of quercetin in Dayak onions and analyze the performance of the method of determining quercetin in Dayak onions using the High Performance Liquid Chromatography (HPLC) method. The extraction process was carried out by maceration method, determination of wavelength using a spectrophotometer and determination of quercetin content using HPLC with Aquades pH 4: Acetonitrile LiChrsolv (60:40) mobile phase. At a wavelength of 370 nm, quercetin appears in a time span of 8-9 minutes. It was found that the ethanol extract of the Dayak onion had more types of secondary metabolites than the Dayak onion with HCL extract. A total of 2.13 grams of ethanol Dayak onion extract was obtained after mixing and 1.09 grams of HCL Dayak onion extract was obtained after fractionation. The total average content of quercetin in ethanol Dayak onions was 4.46% and Dayak onions HCL 1.47%. The standard deviation value is 17.9555 %, the relative standard deviation with value of 2.4127%, for the detection limit of 11.7107 ppm, and the quantization limit of 39.0355 ppm. It can be concluded that the ethanol Dayak onion and HCL Dayak onion contain quercetin in the 8-9 minute range. Keywords : Onion Dayak, Quercetin, HPLC
