UJI AKTIVITAS ANTIOKSIDAN EKSTRAK KUNYIT KUNING (Curcuma longa L.), KUNYIT PUTIH (Curcuma zedoaria) DAN KUNYIT HITAM (Curcuma aeruginosa Robx.) DENGAN MENGGUNAKAN METODE 1,1-DIFENIL-2- PIKRILHIDRAZIL (DPPH)

ANTIOXIDANT ACTIVITY TEST ON YELLOW TURMERIC (Curcuma longa L.), WHITE TURMERIC (Curcuma zedoaria), AND BLACK TURMERIC (Curcuma aeruginosa Robx.) EXTRACT USING 1,1-DIPHENYL-2-PICRYLHYDRAZYL (DPPH) METHOD Name: GRACE SABATINA NIM: 1541007 ABSTRACT A study on the antioxidant activity had been tested on yellow turmeric (Curcuma longa L.), white turmeric (Curcuma zedoaria) and black turmeric (Curcuma aeruginosa Robx.) using free radical 1,1-diphenyl-2-picrylhydrazyl (DPPH) method. Simplicia is extracted by cold maceration method using methanol, dichloromethane and hexane as the solvent. The concentration of the extract used was 12.5 ppm, 25 ppm, 50 ppm, 100 ppm, 200 ppm, 400 ppm and 800 ppm. Measurements of antioxidant activity (inhibition percentage) were carried out using spectrophotometric at a wavelength of 517 nm on a mixture of extract and DPPH solution, then the data were analyzed using ANOVA statistics and Duncan's Multiple Range Test. Phytochemical screening results showed that the three types of turmeric had tannins, flavonoids, phenolates, monoterpenes, sesquiterpenes, quinones and saponins, whereas steroids and triterpenoids were only possessed by yellow turmeric and alkaloids possessed by white and black turmeric. The results showed that the extract of the three types of turmeric give a significant difference in the antioxidant activity (p = 0,000), where yellow turmeric provided the best antioxidant activity with an average of 54.63%. Based on the type of solvent used, the results showed that dichloromethane was the best solvent (p = 0,000) which give antioxidant activity with an average of 46.66%. The increase in extract concentration from 12.5 ppm to 800 ppm is in line with the increase in the antioxidant activity, which is indicated by the statistical results (p = 0,000). Furthermore, identification of antioxidant compounds that involve in DPPH free radical inhibition can be carried out, as well as pharmacological testing.
