ABSTRACT STUDY OF ANTIOXIDANT ACTIVITY OF CAFFIR LIME LEAVES EXTRACT (CITRUS HYSTRIX DC ) USING METHOD 1,1-DIPHENYL-2-PICRILHYDRAZIL (DPPH) Rentauli Silaen NIM : 1141010 Antioxidants are electron donating compounds which are able to prevent or to decrease oxidative damage of the body. In search of natural antioxidant compounds, this study of antioxidant activity assay of ethanolic extract, dichloromethane fraction, and acid fraction caffir lime leaves (Citrus hystrix DC) with DPPH is being done. From the study of active compounds, it was found that caffir lime leaves contain chemical compounds such as flavonoids, alkaloids, steroids, saponins, tannins, and triterpenoids. The yield of concentrated ethanol extract of caffir lime leaves simplicia obtained is 2.50 %, or as much as 16.30 grams from 300 grams of simplicia. Fractionation from ethanolic extract was performed using dichloromethane and acid solutions, the yield is 1,72 % of acidic fraction and 1,41 % of dichloromethane fraction. Results of ANOVA analysis with the time of mixing with DPPH for ethanol extract, dichloromethane fraction, and acid fraction showed that p = 0.000 which is lesser than α = 0,05. The results showed that the antioxidant activity of the ethanol extract, dichloromethane fraction, and acid fractions of concentrated caffir lime leaves after being mixed with DPPH show that the highest antioxidant activity is at concentration of 500 and 1000 ppm. The analysis showed that the dose variation influence on the amount of antioxidant activity. This is confirmed through the regression analysis that showing that increasing the dose of the extract increases the antioxidant activity: the ethanol extract by 77,4%, dichloromethane fraction by 87,7%, and acid fraction by 83,00%.
