AFINITAS PENAMBATAN ISOLAT FLAVONOID TANAMAN ALPUKAT (Persea americana) PADA RESEPTOR NF-kβ, COX-1, dan COX-2

ABSTRACT AFFINITY OF DOCKING ISOLATE FLAVONOID FROM AVOCADO (Persea americana) ON RECEPTOR NF-kβ, COX-1 and COX-2 Brenda Christy Januarita Siringoringo NIM: 1241011 Research about anti-inflammatory activity is still found interesting in new drugs development. As a target of anti-inflammatory drugs, receptors such as NFkβ, COX-1, and COX-2 can be simulated by molecular docking with various Flavonoids from other medicinal plants. In this study, docking simulation of 13 isolates Flavonoids from Avocado and used Aspirin isolate for comparison. These 14 isolates used as ligands on NF-kβ, COX-1, and COX-2 receptors had done. Those isolates downloaded from National Center for Biotechnology Information (NCBI) which is expanded by Pubchem. Test parameter in this study is binding affinity that obtained using PyRx that equipped with Autodock Vina. Results data has been analyzed with Two-Way ANOVA statistics and showed that (1) There is an effect of isolated Flavonoid from Avocado to NF-kβ, COX-1, and COX-2 receptors (p = 0,000) with Catechin showed highest binding affinity (-8,1000 kcal/mol) which means Catechin has higher binding affinity than Aspirin (-5,2837 kcal/mol). (2) There is a differences of effect of NF-kβ, COX-1, and COX-2 receptors to isolates Flavonoids of Avocado (p = 0,000) with COX-1 showed highest binding affinity (-7,9969 kcal/mol). (3) Interaction between isolates Flavonoids of Avocado and NF-kβ, COX-1, and COX-2 receptors effected the binding affinity (p = 0,030).
