PENGARUH KONSENTRASI ISOSORBID DINITRAT (ISDN) DAN NATRIUM NITRAT (NaNO3) DALAM KONVERSI NITRAT (NO3) MENJADI NITRIT (NO2) PADA BAKTERI Escherichia coli

ABSTRACT This research has canied out with the tiile 'THE INFLUENCE of /SOSORE,DE DIN|TRATE (SDN) and SODTUM NITRATE (NaNq) CONCENTRATTON in the CONVERTTON of NITRATE (N0€) TO NITRITE (NO) by Escherichia coli". The purpose of this research is to determine whether there is the influence of batcteria in the human digestive sysfem fo the decomposition of nitrate into nitite by in vitro using Escherbhia coli on two treatments, by providing ISDN (lsosorbide Dinrtnte) and NaNOs (Sodium Nitrate). This research is an experimental laboratory with vaiance group according to growth medium, which is contol group and 6 teatment groups that is given ISDN and NaNOs with different concentration. On each group bacteria is added according to McFarland 0.5 standard, and followed by addition ot 3OO0 ppm glucose solution, with equal volume. After 24 hours incubated, it is continued by measuing the growth medium by using a spectrophotometer to see the absorbance in the solution that is used to measure the number bacteia. By using fhe Griess method, the nitrite concentration in every sample can be measured. The changes of color into red represent nitrite that is formed. The resuft of absorbance measurement is catibrated with the catibration cuNe that is obtained before, for the number of bacteria Y=2,782 x and for the nitrite concentration Y=0,0278 x + 0,053. The resuft of measurement of the number of bacteria and nitrite concentntion is statistically analyzed by using the one way ANAVA. From the result, the conclusion is obtained that there is no influence of the initial concentration of nitrite in growth media on the number of the Escherichia coli Ho1) wrth q=0.044. The second research conclusion showed there is the inftuence of the initial concentration of nitrate in the growth medium to the amount of nitite formed by the Escherichia coli (Ho2) with q=0.000. The resuft of Duncan test for the second conclusion showed that the growth medium which is 1OO0 ppm ISDN with addition ot 3000 ppm g/ucose solution was the group that give the most influence against the conversion of nitnte into nitrite by Escherichia coli. The second highest group of Duncan analysis are the |OOO ppm NaNOsand ISDN 500 ppm group, while the other group left did not give sig nificant different.
